Biological mixtures comprised of multiple individuals’ tissues or bodily fluids are commonly encountered and gathered during crime scene evidence collection. Sexual assault samples are one such mixture, usually comprised of epithelial cells and sperm. Sexual assault mixtures may contain a large amount of non-sperm cells in comparison to sperm cells. In such instances, it is imperative to separate the DNA from each cell type and to optimize the yield of DNA from the sperm fraction by use of a differential extraction. This study set out to discern the effects Proteinase K concentration, SDS concentration, incubation duration and temperature have on the differential extraction efficiencies and premature spermatozoa lysis of aged/dehydrated mixed cell samples. These results were compared to a pristine, unaged/not dehydrated sample set extracted under standard differential extraction conditions. It iv was discovered that under all conditions tested, complete epithelial cell lysis was accomplished. All aged/dehydrated samples not only yielded smaller DNA recovery than unaged/not dehydrated samples, but also contained significant sperm cell prelysis when compared to pristine samples. Variations in the differential extraction method showed no significant difference in the level of male DNA in the non-sperm fraction for all conditions tested, suggesting that the differential extraction procedure is robust and early lysis of sperm seems to be sample dependent and not lysis condition dependent.
